چکیده

Background TSLP has been described as a “master TH2 cytokine” with pleiotropic effects. Recent research shows that TSLP can directly activate mast cells and CB CD34 hemopoietic progenitor cells (HPC), with release of TH2 cytokines/chemokines in a TSLP-dependent manner [1,2]. Additionally, TSLP may modulate the function of CD34 cells via changes in hemopoietic cytokine receptors (HCR), which are altered in CB CD34 cells of atopic at-risk infants [3]. In this study, we aimed to characterize HCR expression on CD34 cells and to examine CD34 cell differentiation following TSLP stimulation.

The effects of poly I:C stimulation of primary bronchial epithelial cells and TSLP secretion on CD34+ progenitor cell eosinophil and basophil differentiation

Background In asthmatic lungs, elevated levels of thymic stromal lymphopoetin (TSLP) are linked to eosinophilic inflammation and disease severity [1]. TSLP is involved in initiating a TH2 inflammatory response, through activation of T cells, and recently, CD34+ hemopoietic progenitor cells [2,3]. However, the biological effects of epithelial-derived TSLP on human peripheral blood (PB) CD34+ pro...

The effects of thymic stromal lymphopoietin and IL-3 on human eosinophil–basophil lineage commitment: Relevance to atopic sensitization

An important immunopathological hallmark of allergic disease is tissue eosinophilic and basophilic inflammation, a phenomenon which originates from hemopoietic progenitors (HP). The fate of HP is determined by local inflammatory cytokines that permit "in situ hemopoiesis," which leads to the accumulation of eosinophils and basophils (Eo/B). Given that recent evidence supports a critical immunom...

Regulation of Thymic Stromal Lymphopoietin (TSLP) receptor expression

Objective/purpose TSLP plays a major role in the induction and effector phases of allergic diseases by acting on dendritic cells, mast cells (MCs), T cells and CD34 hemopoietic progenitor cells. Whereas the cellular origin and the mechanisms regulating TSLP production are well documented, little is known about the regulation of TSLP receptor expression. We analyzed the regulation of TSLP-R with...

Thymic stromal lymphopoietin promotes human eosinophil-basophil lineage commitment: a key role for tumor necrosis factor-alpha

Background Allergic diseases are characterized by tissue eosinophilic and basophilic inflammation. Both epithelial-derived thymic stromal lymphopoietin (TSLP) and eosinophil/ basophil (Eo/B) lineage-committed progenitor cells are upregulated and found at sites of allergic inflammation [1-3]. We have previously shown that TSLP mediates the differentiation of peripheral blood (PB) CD34 progenitor...

A New Two Step Induction Protocol for Neural Differentiation of Human Umbilical Cord Blood-Derived Mesenchymal Stem Cells

Background: In this study, we examined a new two step induction protocol for improving the differentiation of human umbilical cord blood-derived mesenchymal stem cells into neural progenitor cells. Materials and Methods: Human umbilical cord blood-derived mesenchymal stem cells were first cultured in Dulbecco’s modified eagle medium supplemented with 10% fetal bovine serum in a humidified incu...
